Our company now has 200 square meters of space for breeding zebrafish; 3 sets of independent cultivation system carrying 100 000 zebrafish breeding; molecular laboratory; micromanipulation laboratory and fluorescence imaging laboratory, and can quickly make different lines and the related studies. So far, we have signed the contracts with the Chinese Academy of Sciences, universities and hospitals and other institutions for developing different zebrafish models which are used for basic research, disease and drug development. 斑马鱼最早在20世纪30年代就被用于环境检测研究,其正式作为一种模式生物应用于科学研究则始于1972年,这一年美国俄勒冈大学George Streisinge 教授开始斑马鱼发育生物学研究和模式动物建立工作。相较于小鼠模型,斑马鱼模型极大的填补了小鼠模型的局限性造成的空缺。斑马鱼是目前唯一适于高通量药物筛选的脊椎类动物,并拥有个体小、发育周期短 (24小时器官便可形成)、身体透明(可直接观察内部器官)、单次产卵数较高(150~200 枚)等特点。
斑马鱼有着其不可比拟的优势:
1.表型直观
斑马鱼与小鼠模型相比,其身体透明,能在显微镜下进行直观的单细胞表型观察。小鼠模型通常为组织水平研究,通过观察脏器变化来进行观测,而斑马鱼模型可以达到细胞水平的研究,在移植异源细胞后可直接在显微镜下观测到斑马鱼体中异源细胞的迁移等情况。
2.研究周期短
斑马鱼早期生长发育非常快,可大大缩短实验周期,从而加快实验进程。在斑马鱼幼鱼中移植异源细胞后,可在2-4天内通过显微镜观测和评估异源细胞的增殖和迁移等参数。
3.成本低
与小鼠模型相比,斑马鱼所需的养殖空间更小,饲养成本更低,所需要的管理人员更少(仅1人即可养殖和管理上万条斑马鱼)。利用斑马鱼开展研究时所消耗的实验材料更少,尤其适合整体动物上开展规模化的筛选研究工作。
斑马鱼最新的文献报道了prkdc-/-, il2rga-/-斑马鱼(缺乏T、B及NK细胞),可以在37℃时移植多种人类肿瘤细胞,并能使单个移植细胞动态可视化,例如,追踪人类横纹肌肉瘤(一种小儿肌肉癌)的迁移和增殖状态,并通过实验确定组合olaparib PARP抑制剂和替莫唑胺DNA损伤剂的临床前功效,验证横纹肌肉瘤可视化治疗反应。这样我们就可以将患者来源的细胞移植入斑马鱼模型中,为将来开发个性化治疗方法开辟新的途径。
